چکیده

Purpose Intratumoral immune activation can induce systemic immunity and anti-tumor activity. Imiquimod is a creamformulated, TLR-7 agonist that is FDA-approved for the treatment of non-melanoma skin cancers, but has limited activity against melanoma. In the current study, we studied the anti-tumor activity and mechanism of action of a novel injectable TLR 7/8 dual agonist, 3M-052, which remains at the site of injection to avoid systemic distribution.

Intratumoral immunotherapy with the TLR7/8 agonist 3M-052

TLR agonists have demonstrated anti-tumor activity in both animals and humans. Clinically, successful antitumor activity with TLR agonists was most often observed when the molecules were applied locally to dermatological cancers. This mode of administration was also associated with fewer and less severe adverse events (AEs) when compared to systemic administration. The efficacy of TLR agonists ...

Induction of potent systemic anti-melanoma immunity through intratumoral CD40 activation and checkpoint blockade

Purpose Agonistic CD40 antibodies generate strong tumor specific CD8 T cell response and anti-tumor activity; however systemic anti-CD40 therapy has been associated with cytokine release syndrome and liver toxicity. We studied the anti-melanoma activity and mechanism of action of a recombinant adenovirus expressing a stabilized version of CD40L (rAd.CD40L) by local intratumoral delivery approac...

Microenvironment and Immunology Localized Immunotherapy via Liposome-Anchored Anti- CD137 þ IL-2 Prevents Lethal Toxicity and Elicits Local and Systemic Antitumor Immunity

Immunostimulatory agonists such as anti-CD137 and interleukin (IL)-2 have elicited potent antitumor immune responses in preclinical studies, but their clinical use is limited by inflammatory toxicities that result upon systemic administration. We hypothesized that by rigorously restricting the biodistribution of immunotherapeutic agents to a locally accessible lesion and draining lymph node(s),...

TLR agonist rHP-NAP as an Adjuvant of Dendritic Cell-Based Vaccine to Enhance Anti-Melanoma Response

Background: Melanoma is a common and malignant cutaneous tumor, which is responsible for a large proportion of skin cancer deaths. Dendritic cell (DC)-based vaccines have achieved positive results in the treatment of melanoma because of their ability to induce cytotoxic response to facilitate tumor elimination. Objective: To improve the efficacy of dendritic ce...

TLR Agonists for Cancer Immunotherapy: Tipping the Balance between the Immune Stimulatory and Inhibitory Effects

The agonists of toll-like receptors (TLRs) have been actively pursued for their antitumor potentials, either as monotherapy or as adjuvants to vaccination or other therapeutic modalities (1). A search on ClinicalTrials.gov using the key words “TLR” and “cancer” returned 34 listings. The idea of using TLR agonist to provide a “danger signal” and break tolerance to tumor antigens has been well em...
